I want to set a calibration curve for SRB using uv-vis machine. I've read many papers stating that they dissolve SRB in presence of an additional compound such as TiO2. Do I have to do the same? Can't I just dissolve it in de-ionized water and detect it?

If it is soluble in water, sure you can just dissolve in DI water an make the calibration.
I also would believe you mean sulfate reducing bacteria. Bacteria are not soluble, only dispersible, if you do not use drastic conditions. Use of 'optical density' OD (better extinction) is a common method in quantifying cell concentration. No idea about using TiO2, or what is your SRB, and what articles you are referring to?
Michael is correct on all terms, I was too lazy to google SRB, but certainly using abbreviations without introducing them should be avoided
Yea, I should've noted that I meant Sulforhodamine B monosodium salt (ie) SRB dye.
