I have done simultaneous treatment of two drugs in in vitro studies. Now I have to do pre-treatment studies. In simultaneous treatment I have given both the drugs and check for viability after 24 hours. However I have some doubts in designing pre-treatment studies. Is it scientific to treat cells with one drug (the protective drug) for 24 hours followed by the another for 24 hours to get the results of combination group? Also in case of drug controls how much time I have to treat the cells with each drug?
Hi Abhilash! Nothing wrong with pretreating your cells with a first drug for 24h and then apply your second drug and assess its effect. However, you might have to figure out how long you have to pretreat cells to be efficient - maybe you could perform a time-effect response to make sure you are playing with the right timing. As for drug control you have to use exactly the same timing as for your test drug.

It's all OK with pretreatment, however you have be sure that 24 h is an optimal time. Also I would check the pharmakokinetics of both drugs. Controls should be the same time treated as the certan drug was used in the combination

in our experiments we put the cells (T cells incubated with the dendritic cells for 7 days + drug) or (pbmcs with drug for 5 days)
after this time the cells should be ready for an assay
