I just started working with human aortic smooth muscle cells (HASMC) for the first time and experienced contamination problems. I purchased Lonza's human smooth muscle cell systems with their media and the cells arrived to be in P3, from a 21 year old donor, and packaged in August, 2014. Is it normal for such cells to be well in its passages when they arrive? I received cells in P0/1 most of the time so this was new for me. Also, is this cell line usually tricky/sensitive to work with? Any tips on culturing these cells would be great!
P3 is fine in case of ASMCs.
If you got contamination when you cultured the P3, then you may ask for another batch of cells.
Did you add P/S in your medium?
Yes, human ASMCs are more difficult to culture than the rat or rabbit ASMCs.
