Dear All,
Can someone help me in the determination of these enzyme activities in the barley leaves for example; Catalase (CAT), Guaiacol peroxidase (GPX), SOD and Polyphenol oxidase (PPO) activities.
1. GPX activity was assayed following the method of Chance and Maehly
(1955).
2. activity of APX was measured according to Nakano and Asada (1981).
Please explain this sentence:
{One unit (U) of peroxidase activity was represented by the amount of enzyme catalysing the oxidation of l mol of guaiacol in 1 min and the enzyme activity was expressed as U g−1 FW}
3. SOD activity was measured following the method of Beyer and Fridovich
(1987).
{What is this ? 1 U of SOD is defined as the amount required for inhibiting the photoreduction of NBT by 50%.}
and I also need to express as:
{ Enzyme activity was expressed as change in OD/min/g fresh weight of leaf}
Here are OD values of my treatments
.154 for 0 minute, .201 for 30 sec, 1 min for .239, 1:30 min for .271, 2 min for .306, 2:30 min for .339, and 3 min for .370.
