The absorbance value (A) of the sample can be calculated using the following equation:
A = (A520nm-A700nm) pH 1.0 - (A520nm-A700nm) pH 4.5
Total Monomeric Anthocyanin Content can be calculated using the following equation:
TAC= (A ×MW × DF ×1000)/ (Ƹ×𝑙)
Total Monomeric Anthocyanin Content can be calculated as mg of cyanidin-3-O-glucoside (C3G) per L of sample using a molar extinction coefficient (Ƹ) 26,900 Lcm-1mol-1, molecular weight (MW) 449.2 gmol-1 for C3G, path length (l) in cm and the appropriate dilution factor (DF).
Please find the attached file to calculate the total anthocyanin content. However it basically calculated the TAC based on malvidin-3-O-glucoside, but you can to do the same way to find your answer.
Hi Terun, The reason for using Cy3G as external standard is that it is the main anthocyanin component in your samples. If it is not, let's say pelergonidin-3-glucoside (Pg3G) is the main anthocyanin in you sample. Pg3G should be used as Pg3G equivalents in you calculation.
