I would be interested to hear if anyone has applied NanoDSF (as performed by the Prometheus instrument from Nanotemper) to study the unfolding of membrane proteins reconstituted into liposomes or nanodiscs. Is this theoretically possible?
Theorically is possible. I've not used the Prometheus instrument for this application, but the system just measure the fluorescence of the aromatic residues of the protein. This measure has been used to study the unfolding of membrane proteins reconstituted into liposomes. I've performed myself this kind of experiments with liposomes, but in a conventional spectrofluorimeter, and works well. There is no reason Prometheus should not work too.
