I have come through a couple of publications claiming that simple MTT could replace the Lactate Dehydrogenase Assay in malaria drug susceptibility testing. MTT is quite cheap compared to LDH kit, but I am a little confused, given the non specificity of MTT, as compared to LDH. Could somebody help clarify this ?
MTT acts on the mitochondrial enzymes. And since the plasmodium mitochondria, i.e. apicoplasts are not well developed, it is advised not to use MTT.

I have successfully used MTT to detect the presence of Plasmodium ookinetes in ookinete binding assays in this paper (see attached).

LDH kit would do much better than MTT since the antigen does not persist in the blood following successful treatment i.e. it clears from the blood once the parasites are eliminated. MTT measures viability of cells and therefore its reduction occur in a medium with metabolically active cells.The implication is that it would take much more time to determine effectiveness of the drug.
ready to use LDH kits are available for malaria detn. Also LDH based ELISA was used earlier for quantification.
As advised by Simon, MTT production was linearly and directly proportional
to the number of ookinetes seeded into microplate wells. U could giveit a try
