Could someone give any suggestion about possible mechanism of miRNA transcription? The problem is: 1) miRNA-X is expressed in cardiac tissue and 2) miRNA-X is encoded in intron (length – 93.5 kb) of Y-gene coding region, which is highly expressed in brain, but not in a heart (we didn’t try yet primers for different Y-gene regions e.g. different splice variants or introns). There are reports though that untranslated region of that gene is expressed in non-neuronal cells, but miRNA-X is in coding region. So my main question is: can miR be expressed independently of the host gene? Should we start testing our miRNA-X expression regulation from measuring expression of different regions of the host gene?
