Could someone give any suggestion about possible mechanism of miRNA transcription? The problem is: 1) miRNA-X is expressed in cardiac tissue and 2) miRNA-X is encoded in intron (length – 93.5 kb) of Y-gene coding region, which is highly expressed in brain, but not in a heart (we didn’t try yet primers for different Y-gene regions e.g. different splice variants or introns). There are reports though that untranslated region of that gene is expressed in non-neuronal cells, but miRNA-X is in coding region. So my main question is: can miR be expressed independently of the host gene? Should we start testing our miRNA-X expression regulation from measuring expression of different regions of the host gene?
Hi Sergey, According to Ozsolak et al. 2008. Genes & Dev., intronic miRNA might have independent promoter for faster transcription and are likely located far from host TSS.
Dear Seregy, miRNAs does not need host gene promotors as shown by many authors (http://www.ncbi.nlm.nih.gov/pubmed/21857109). From our reserch we have found that miRNAs can originate from any where in genome.
In your case miRNA is coming from intronic region which can be a miRtron. For further knowledge on miRtrons also please read this papar "http://www.ncbi.nlm.nih.gov/pubmed/22955976" where several miRtrons have been identified.
