I have did two transfection with pcDNA3.1 which contains my gene constructs separately and they are working fine. Next, I am planning to express two genes in a marine cell, however, both genes are constructed in the same plasmid (pcDNA3.1) because I only have pcDNA3.1. Can I co-transfect these genes with the same plasmid? For example, pcDNA3.1-A and pcDNA3.1-B co-transfection in a marine cell. Will plasmid incompatible occurs between two pcDNA3.1?

The short answer is YES. Both genes are likely to be expressed equally if the two plasmid preparations are of comparable quality and both plasmids are transfected into cells in 1:1 ratio. However, levels of two expressed mRNA and proteins in the transfected cells could vary due to differential post-transcriptional regulation of the encoded mRNAs and corresponding proteins.
