Is there anyone who is using FlexMap 3D system for multi-analyte ELISA? For multiplex cytokine measurement at one time, Luminex technology has been used. The most well-known machine is Luminex 200 or Bio-Plex 200, which are based on a 96 well platform. Because of the quite high cost of 96 well tests for multianalytes, it would be helpful if a 384 well platform of FlexMap 3D machine is used. I’d like to know if the 384 well platform can reduce the reaction mixture usage compared to the 96 well platform. And if possible, I want to know how much I can save it.

By reagents you mean the beads? You need to think about what this system is. It depends on getting a statistically valid sample of beads. So the counts of beads should be the same in the 96 well and 384 well systems. The 384 well system just allows you to run more assays simultaneously.
If you haven't done 384 well plates before, it's not something to try without a robot. If you try to do it manually, you are virtually certain to spill material into neighboring wells. I found that when I did an (unpublished) study of 96 well plates with 6 people in the lab. All of them were sure they had not done it, and all of them had, but to varying degrees using a single pipetter and two rows on a plate. It was a simple test. I just used water, and then used an RIA plate and took each well out and weighed it.
If you want a more thorough understanding of the system, here's my thesis. The Luminex system is remarkably good, and the best multiplexes take most of these factors into account. Since this was published, I understand that Luminex has gotten better with its electronics.

If you tell me what the assay is, and how many multiplexes there are, I can comment better perhaps. But please read the thesis to get a better understanding.
