I'm looking to try this in place of normal goat serum for a wisteria floribunda lectin stain. In my IF protocol, 10% of the final volume of my blocking solution is goat serum. If I were to go the carbo-free route, could I just use this solution in place of normal goat serum? For example, if I needed 5mL of goat serum, could I just use 5mL of carbo-free?

Carbo-free blocking solution is usually recommended for lectin staining because it is a protein-based agent that does not contain glycoproteins.

Our usual protocol for the IF blocking solution was goat serum and donkey serum. I replaced all of the serums with a carbo-free blocking solution for my lectin staining, and it worked really well (except some lectins that show higher background, depends on the tissue you are staining, I guess). WFA works for me in the brain using the carbo-free blocking solution. You can replace the same amount of serum with a carbo-free blocking solution.
Lalhaba Oinam, thanks! I actually diluted the carbo-free solution down to 1x, added 5% triton-x (10% stock solution) and my stain looked good. I may repilot the stain by doing what you suggested.
Do you add triton-x or anything else to your blocking solution?

does anyone know if using the carbo-free blocking solution is good to stain nuclear cytokines?
I am getting very low signals.
I block the sections with 1x carbo free (made in 1xPBS + 0.1% tween).
