Can I transfect my cancer cells with shRNA twice?

My gastric cancer cells have already been transfected once with shRNA and gene knockdown has been verified. After a period of time in culture, the behavior of the cells changes. Can I re-knockdown this batch of cells? Or must use the WT cells to knock down and validated again?

It would be best if you started from the WT cells. If you need a durable knockdown with your shRNA, switch to a lentiviral system rather than using plasmid transfection.
