I took four slides out from -80 freezer. Then I realized one of the reagents was over but I couldn't see it before because the container is dark. What can I do? Do another staining or can I storage them back?
Yes, if they are already fixed and stained. We repeatedly take them in and out of the -80. For us, the freezer is helping to keep the fluorophores or stains from degrading, and the freeze/thaw does not have a negative effect on anything.
But if your slides are fresh tissue that has not been fixed, you may have problems.
my experience is with 'fresh' muscle sections on slides that once they have been thawed and refrozen that staining doesn't work as nicely. Best is if they're already thawed, just do another staining with the thawed slides in order not to waste them. At least with 'fresh' muscle, -20C is sufficient for storage of sectioned material.
Yes if it is fixed you can restore them. If they are frozen sections there 100% grantee to say you cannot stain them again , but they might not be perfect. Since you have taken them out you can go for try and find what will happen in such situation.
