I have prepared samples in SDS loading buffer to load the gels 5 days back. Can I still use the same samples which are already prepared to repeat the western blot because I don't have enough samples to prepare fresh loading samples.
No problem at all and you can use the samples again. Protein degradation is not proportional for all samples. Thus as long you see the same bands for all samples at the same hight than you can be sure about integrity of protein that you want to detect
I lyse my cells in SDS buffer and boil for 5 mins. I then store at -20 degrees. The sample is then boiled for a few minutes again just prior to loading onto a gel, the remaining sample goes back in -20 freezer.. You can do this 4 or 5 times before the repeated boiling will start to affect your sample.
