I diluted my primary monoclonal antibody (phospho PKR) in 2% BSA and incubated my membrane overnight. In the end, the membrane resulted in very high background and now I realized that milk (specifically the phospho protein contents) may interfere with binding. Nevertheless, I re-use and recycle my primary antibodies. My question is, can I still use my recycled primary phospho antibody even though I blocked my membrane with milk? The antibody was diluted with 5% BSA and TBST 1X? Did the milk in the membrane somehow made my antibody non-functional?
