I am planning to work with CACO-2 cell line for drug permeability assays in which transwells are used. Can anyone please tell me which microscope should I use to follow the cell growth and morphology until it reaches full differentiation (because the hight of the transwell and the membrane on the insert might interrupt the light..)? Which membrane of insert is recommended for better growth of the cells and for better imaging - PET or Polycarbonate? also, I have read a few protocols in the literature saying CACO-2 cell can be differentiated for only 3 days after transferring to transwell. Did anyone tried it and succeed?
