I have extracted collagen and tried estimating total protein content of the same using Lowry's, Bradford's and Biuret's methods. All of them gave low values and I realised these methods will not work well for collagen. I found modified Lowry's method and modified Bradford's method for collagen. But, will these methods work well for BSA too as I shall be using it as standard to determine total protein content of collagen. Has anyone tried the modified method?

Do read the paper about the modified Bradford assay carefully (see enclosed file and especially look at Figure 3). Adding 0.0035% SDS leads to the conclusion, and I quote “This simple modification in the preparation of the reagent for the Bradford assay allows similar response curves to be obtained for collagen and non-collagen proteins, making the modified assay of potential use for protein determination in collagen-rich samples such as pancreatic extracts.”
