I have noticed various authors use different voltage and time while running their gel electrophoresis such as 100V for 30 min and 75V for 35 min etc.
Can anyone kindly suggest a preferred condition so i can obtain good bands.
I am using 1% agarose with TAE buffer stained with midorin green

The voltage recommendation for agarose gel depends on several parameters including, the Agarose type, electrophoresis buffer and the gel apparatus. The voltage applied is determined based on the distance between the electrodes, not on the gel length. The general recommended voltage are:
5-10 V/cm for DNA size

Normally, if you run your gel at lower voltages (70-80 V), your bands will be clearer than if you run it at higher voltages. If you are running a DNA ladder with your samples, run the gel until the DNA ladder bands of interest (those corresponding to the size of your protein) are enough separated to allow for determination of the size of your sample bands. Alternatively, if you are using a loading buffer with Coomasie blue, run the gel until the dye line has almost reached the bottom of the gel.

the common rule for nucleic acid electrophoresis is to apply a voltage of 1-5 V/cm (measured as a distance between electrodes). Running time must be adapted to the size of the fragments you are resolving and the lenght of the gel.

If you want a good band resolution, i advise to use low voltage at longer duration. Especially if you use TAE which is easier to get hot compared to TBE. As for duration, usually you can use loading buffer movement as a marker, since some loading buffer represent movement of DNA in size of several hundred base pairs.

In our Lab we run several type of bacterial PCR product and we use 90V for 30min. Works fine for all.
