We have SV40 immortalized white adipose tissue-derived preadipocyte cell lines, but these cell lines can not do differentiation. hTERT might be another way to get cell lines if it will not impact preadipocyte differentiation.
We have successfully immortalized inguinal WAT pre-adipocytes with SV40, but this needs to be done in cells from newborn mice. The use of hTERT or combinations of SV40/hTERT has been described in the literature to immortalize pre-adipocytes.
Thanks Emilio, we also successfully immortalized inguinal WAT pre-adipocytes with SV40 from young mice, but, these cells can not differentiation after a few passages. Do you have similar evidence that SV40 immortalized inguinal WAT pre-adipocytes can not differentiation. We also did cell lines from inguinal WAT pre-adipocytes after 3T3 protocol and single cell clone, unfortunately, our interesting gene has different role in differentiation using primary inguinal WAT pre-adipocytes and cell lines. We just need a fast way to get cell lines except SV40 without impact differentiation.
