This can mean that the cell population secreting PSA is destroyed or that the cell's ability to secrete PSA is blocked. Does anyone know a study showing the response of a prostate cancer cell culture to hormone blockade?
There are numerous studies (starting from early 1990), using androgen responsive LNCaP prostate cancer cell line, demonstrating that androgen withdrawal in this prostate cancer cell line results in inhibition of cell growth. Further, a continuous culture of LNCaP cells in androgen-free cell culture medium in due course results in selection of cell clones that are androgen sex hormone-independent. These cell variants are resistant to cell death by apoptosis.
There are a lot of studies ,that shows, androgen responsive prostate cancer cell lines responded androgen withdrawal with inhibition of this type of cell growth. In fact the medium then will be covered by androgen independent cell clones which are resistant to androgen withdrawal.
I am aware of some studies using LNCaP cell lines showing INHIBITION OF CELL GROWTH. The future of these cells is uncertain. Corresponding to my observation ADT is followed by
1. reversible reduction of PSA-secretion followed by a recovery of these cells.
2. irreversible reduction of PSA-secretion possibly as a consequence of the extinction of this cell line.
To date I was unable to find evidence that the cell line was starved out or was able to restart cell division.
Interpreting Atif Mahmut Akdas comment using ADT would select castration resistant cell clones. This would mean that androgen sensible and androgen independent cell lines coexist in the untreated prostate cancer?
Several clinical studies showed histopathogic chenges induced by androgen deprivation: regressive effect and apoptosis sere more pronunced in low grade disease rather thai in high grade. However chenges were reversibile. You can find a recent pubblication on Future Oncology. A medline Search including neo adiuvante hormonal treatment is helpful.
