I accidentally used a contaminated solution for making aliquots for my growth factors. The growth factors are very expensive and we still want to use them. I am sure that the solution I used (0.1%BSA) was contaminated with fungus. The fungus was white and like a ball with long hair.
I store the contaminated growth factors in -20 C. Can someone tell me a way so that I can still use them in my medium and get rid of the contamination? Thank you!
Additionally, I cant really filiter them since there are only 2 ml in total.
Sherry, I hate to break a bad news, but I'm afraid there is nothing that can be done to reverse the situation. You are better off dumping the solutions. For starters, you can't freeze it dead - that's for sure. One could think the filtering with sterilizing filter could help - and it might. The concern in that situation is that the thing might have eaten your growth factors bare and released a ton of toxins in the process - and those things will kill your cells for sure if the fungus itself won't. Not a good way to die. Sorry for being so straightforward... I hope it helps at the end. Good luck!
Sherry: I basically agree with Vadim's opinions, but would consider to save your solutions (more than one growth factor?) if they were frozen immediately after preparation. In that case, you can use a 0.22 micron (not 0.45 micron) disc to filter-sterilize and "wash" or "dilute" with one volume of additional buffer or medium to get the maximal recovery. Good luck
Unfortunately, the only thing you can do is to dump your contaminated cytokines and to start with fresh ones.
