We plan to develop RADseq libraries from damselfly larvae grown in the lab, and in this moment our larvae have a size of approximately 5 mm and weight around 3 mg. However, we don't want to keep them much more time in the aquariums to avoid cannibalism (and reduction of our sample size). We think that by the size they have reached enough DNA can be extracted to build our libraries, which needs 1 ug of genomic DNA per sample, but we are not sure. We are going to use Qiagen-DNeasy Blood & Tissue Kit, however it suggests a sample weight of around 25 mg.

Does someone have already built successful RADseq libraries from these small tissue samples? Or does anyone knows more or less the percentage of DNA recovered from insect samples using this kit?
