Can DAB-H2O2 be used instead of TMB for ELISA?

Infact I plan to coat the plate with tissue lysate. Are there other things one needs to look into? Also should BSA be used while diluting secondary antibody in TBST?

No. DAB is a precipitating substrate. If you want less sensitivity, try ABTS. Or ImmunO4.com has an ABTS replacement. I would use a diluent stabilizer for the secondary.
