I am getting some weird results when transplanting human peripheral blood CD34+ cells into humanised mice. After several weeks, the human cells in the bone marrow are CD45+ indeed, but there is a distinct CD45-negative cell population, which is stained positive for human CD3+. Has anyone come across this issue?
If you are certain of your result (no technical errors), I suggest you include in your stain an anti-mouse CD3 Ab and a different marker for human T lymphocytes (ie. CD2). Furthermore, to improve separation, anti mCD45 and anti hCD45 should be included in the same staining. You should be able to get to a reasonable conclusion by trying the above strategy.
Dear Gabriele, many thanks for your reply. Indeed, we used an anti mCD45, and it was not expressed in those human-CD3+ cells.
An anti-mouse CD3 is a good idea, while a human CD2 will be a last resort.

Hello there！ Recently, I encountered a similar phenomenon. I performed surgeries of varying degrees on mice and observed that in two groups of mice, splenic cells exhibited a CD45-negative phenotype, while CD3, CD4, and CD8 were positive. Interestingly, this phenomenon was not observed in the peripheral blood or bone marrow tissues of these two groups of mice.
