No. The purpose of using Atropine is to demonstrate that it and the class it represents (alkaloids) can be recovered via your extraction system.
Yes, I agree with Bruce; but I think it will be nicer if you can do quantification to specific alkaloid(s) instead of as total alkaloid (with identically authentic standard). I attach a nice publication regarding that matter, Best regards
It might seem that caffeine was not an alkaloid.
Of course I agree with both your views. However it would be ideal to use labeled standard (i.e. with deuterium) for quantitation and recovery studies.
Is there any literature review available for the same,even if caffeine is an alkaloid.

If you would like to use internal standard, you should see what class of alkaloids your plant has?
if your plant has indole alkaloids, you have to use indole alkaloid as internal standard.
