I'm doing gene sequencing in radiation resistance bacteria. Can you tell me what steps I should I follow if discovered a new gene to register this in the gene bank?
Hi Mahmoud, is it actually a new gene or is it a predicted function associated with a new sequence? Generally from what I know, at least for new sequence, is that you need to have the Sequin program (free from NCBI) in order to gather all the Metadata etc. Then I think you just send the new seq file to them. Sorry I cant help more I am afraid. Good luck.
The first question is Which gene bank are you planning to make submission? If this is NCBI, please follow the guidance/protocol for submission (http://www.ncbi.nlm.nih.gov/books/NBK51672/).
1) Fist categorize the type of raw data according to How to submit sequence data section (http://www.ncbi.nlm.nih.gov/guide/howto/submit-sequence-data/). At this point you are required to choose one of the option to classify the sequence (complete gene, incomplete gene, gene section etc.....)
2) After figuring out the class of your data you are expected to choose (SUBMISSION TOOLS
& HELP DOCUMENTS) to be informed about the uploading, final quality check.
3) But please fist create an account for NCBI (MyNCBI) This would be requested from you during the protocol.
4) You would be expected to choose of the submission tools (online/offline submission).
5) You are expected to read gene bank submission terms/policy of course (http://www.ncbi.nlm.nih.gov/WebSub/?tool=genbank)
6) After completing these steps you are required to use your account (MyNCBI) possibly by ( http://www.ncbi.nlm.nih.gov/WebSub/?tool=genbank) tool.
But please do not forget that your data is better to be supported by a publication. NCBI submissions make your data available to public.
In case you still need some info, in my experience - it is easier to prepare your new gene (new DNA sequence of a gene?) off line using the Sequin software (available for download here http://www.ncbi.nlm.nih.gov/Sequin/download/seq_download.html). Once you downloaded, then install/run it in your laptop/desktop computer and follow step-by-step sequence data preparation. When it is finish, you will have a some kind of text file (which is not exactly a text file), you need to send the text file to the admin of NCBI DNA database (here is the address: [email protected]). Within two days at most, you will get the accession number for your gene in the NCBI database. However, your sequence will not be available publicly until ca. 6 months after submission (they need to process your submission and take that much time for it). In the meantime, you may use or cite the gene accession number in your publication). So far I have no problem at all in following the instruction for preparing sequence data for NCBI submission using Sequin. Finally, to my understanding, the DNA databases that are available are cross referenced. If you submit your sequence to NCBI, others searching the gene in EMBL (European) or the Japanese DNA database will eventually run across your sequence too. Good luck...
