Can anyone suggest a protocol for PBMC activation to induce granuloma tb model (in vitro) expression on interleukin 6 by IHC methods??

I am trying to set up a co-culture model with machrophage cell line and PBMCs. I am specifically interested in looking at granuloma fomation + morphology Granuloma Tb such as Giant cell, foam cell (induced by machrophage cell line to granuloma).
