What secondary metabolites are u interested in? We have over 25,000 phytochemicals. If you have no interest in a particular one, I suggest u carryout a phytochemical preliminary screening where u make a crude extract and test for the presence of phytochemicals. Your result will guide you on the particular phytochemical to look out for. For instance if alkaloid is present, u might go a bit further to now find out what type of alkaloid is present because subclass of alkaloids alone is tending to a thousand.
Preliminary screening will also guide u on the standards u might need to run a HPLC analysis.

Secondary metabolites extraction can be done by solvent extraction which are of 2 types.
1. By using polar solvents
2. By using non polar solvents
Based on the metabolite you are interested in u can try different solvents where the yield is maximum, and later proceed with using the best solvent for large scale extractions.
Ethyl acetate, Acetone etc are usually used for crude metabolite extraction.
please specify your secondary metabolite..there are many extraction methods which include. hot water .. cold water ectraction.. also some metabolite are pH sensitive, temperature sensitive, having different boiling pint, melting point, solubility in solvents..
Firstly you should have to select your choice of secondary metabolites from the extract, then go for HPLC analysis and by analyzing the standard of secondary metabolites with the samples you also quantify their concentration in the sample extract.
i need a usefull method to extract DNA from oyster mushroom(pleurotus ostreatus). my question is in solution stage of extraction, solution in TE buffer. the paltes of DNA doesnt solve properly. what part of my work is wrong?
