I can find ELISA kits that say they have monoclonal antibodies that are selective to mature insulin, but I can't find a commercial antibody that will admit to not cross-reacting to C-peptide and proinsulin. Has anyone had any luck with doing this?
Francie: We've used hundreds of Abs from Santa Cruz Biotech and found them almost all good. They have a polyclonal (F-16, sc-26793) and a monoclonal (C-12, sc-377071) anti-Insulin B Abs, both of which used N-peptide (B-chain) of a human origin insulin. My own experience was that a direct coupling of an Ab to HRP would enhance WB as well as ELISA specificity and sensitivity. Good luck.
Hi Ke-Wei Zhao, thank you for your answer! I could not find any information on the datasheet from santa cruz regarding whether or not the antibody cross reacts to C-peptide or the proinsulin forms. Do you have data looking at this? We need to find one that does not cross react with these products, but only with the mature insulin form.
