Can anyone recommend a reliable method for extracting plasmid DNA from E.coli? I want to employ as few resources as possible since many of them are not generally available in our lab.
A classical alkaline lysis method described by Sambrook would be helpful, as it requires basic reagents: Tris, HCl, EDTA, NaOH, SDS, KAc, AcOH, and can be performed without RNAse.
in addition to the classic lysis method, the original boiling miniprep also works quite well and requires few reagents as well.
