We are currently using eBioscience ELISA kit (Ready Set Go) for human IFNg detection and we have no problem so far.

The eBioscience kit looks nice from its description on their website, and it is one of the cheapest; however, the detection range seems rather restricted (4-500 pg/ml) compared to other kits in the market. Has that ever been a problem for you? (In the sense, have you had to repeat a sample just because the IFN-gamma concentration went beyond the standard curve?)

My problem with the Pierce plates was that, when I put the exact same samples simultaneously into two plates of the same batch, I ended up getting different absorbance values at the end of the assay. The difference was more than what could be accounted for by random variation.

If you're familiar with using the FACS, I suggest using the legendplex (CBA method), which is both cheap and convenient protocol (metal beads covered by antibodies, so each bead is doing an elisa, and you look at beads fluorescence by facs) - you can buy only the beads for IFNg if necessary..

Thank you, Philippe. Your suggestion is good. I do use flow cytometry in my work, and at some point, I'd need to use a multiplex assay (I use a luminex 200 system). However, for the present purpose, I am restricted to using an IFN-gamma ELISA.

We've been using the R&D Systems (duoset) and it works very well. If you have limited sample availability, I'd also suggest to use the half-area plates from Costar (cat# 3690) so the final volume would be 25ul. In this case a kit would render up to 40 plates instead the 10 plates (when using conventional Maxisorp plates).

Thank you Ricardo. I have considered the duoset for IFN-gamma, because I have good experience using the duoset with other cytokines in mouse-assays. I may eventually get back to it. However, for the time being, I am looking at a preconfigured assay format. The R&D assay looks good, but it is rather expensive and contains only one 96-well plate. That's why I am still weighing my options.

R&D systems ELISA kit for human IFN-gamma is good to use with quite reproducible results...Worth the money !!!!

I usually use the Human IFN-g  OptEIA ELISA Kit BD biosciences. I never had problems, always worked very well.

Many of us consider the gold standard for singleplex cytokine Elisas to be R & D Systems but many companies offer gamma Elisas. With any company product measuring cytokines, be sure they standardize their kit to WHO international standards

Absolutely, Howard. I have all too often used R&D duosets for various cytokine related assays. But it is also true that they are VERY expensive, and sometimes - under some circumstances - it becomes difficult to justify the expenses, especially in the initial exploratory stages.

I absolutely agree.  R & D is very expensive and that's why I stress the need for standardization against the international standard when considering other sources.

I have  used the eBIOSCIENCE human IFN gamma ELISA ready - SET- go. it worked well. maybe u can try the same and check it out.
