Vectors aren't cheap but have permanently once purchased. Life technologies has a large selection with tags, promoters, etc unless you want to include your own.

That depends on your expression system, tag of interest etc. Based on these you will find appropriate vector and find out who's selling it.

First you must decide in which system you want to express your protein:

Bacteria

Yeast

Mamalian

Insect cells

Plants

Next if you need a tag for detection and/or purifying.

Ask in other labs in your institution, if they can give you an aliquot of an usefull plasmid.

Otherwise Clontech, Invitrogen, NEB, Qiagen, Promega etc sell various plasmids.

It depends on what you are doing downstream - every company has good sets of vectors and cloning materials - Novagen, Life Tech, Promega but each have tools for issues encountered. There is the pET system and then vectors for issues with protein insolubility etc... if you look around you will find something appropriate.

The lifesciences data base is really huge! I need to have a closer look on it.

It really depends on your organism or cells of choice. A good general source is http://www.lgcstandards-atcc.org. Yet if you work with yeast or Drosophila you would contact specialised sources like the DGRC.

You can always check in pubmed who has published something with that plasmid and ask them to send you an alicuote absorbed in a filter paper.

instead buy a plasmid you can apply it from researcher. for example: my friend required a vector and no money to buy it , then she asked from one of the reasercher ( he had worked on the same topic) then the reasercher gave her as a gift.

I mostly buy from Addgene costs 60$ each.

It depends much on which kind of plasmids you need. The cheaper and easier thing is to ask a researcher. If you can't do that then you have several sources available. I bought in the past plasmids from OriGene and Invitrogen. My experience with Invitrogen was terrible: they send me bacteria that instead of one plasmid with a specific cDNA contained dozens of different cDNA ( kind of a library with multiple inserts); I asked for a replacement but I got exactly the same stuff again).

OriGene was instead very reliable and I could use immediately the plasmid they sent; they are quite expensive however.

You can also try clonetech to see if they have what you want. I use addgene, but it seems they do not have what you are looking for. What plasmid are you looking for exactly? If you are going to clone your GOI, then it almost seem to me like you should have quite a few choices. But again, you might be looking for one particular plasmid. Good luck with the expts!!!

New English Bioscience ( neb.com ) has a lot of great plasmids for E. coli work, such as a plethora of pET vectors and derivatives. Invitrogen ( invitrogen.com ), now Life Technologies, has many plasmids to select from for yeast work in P. pastoris, or Baculovirus, and some mammalian ones as well. Both those are great sources that can meet most needs. Lucigen ( lucigen.com ) has some plasmids and focuses a lot on fusion protein constructs such as SUMO, so that's one to definitely keep in mind.

Another great source for both plasmids and genes of interest is Open Biosystems ( openbiosystems.com ). And of course, Addgene.

Agree with Patrick boom. Addgene is one of the best source. you can share your own plasmid also you can purchase your desire plasmid. i myself purchased plasmid from addegene 68$ us dollar. for transfection of HEK293 cell line. let me know if you need any help.

Aside from Addgene (which I have also used) have you tried doing a literature search for the type of plasmid you are looking for? You may find a similar plasmid for what you need just by asking other labs to provide it for you (assuming they've published it). There may be some paperwork to process between labs, such as a material transfer agreement, but I've had success just by emailing the corresponding authors. It's worth a shot if you can't find it anywhere else.

Addgene, Promega, Clontech are good choices to try. However, I recommend you to look for your plasmid through other publications. Some times you can easily request the plasmids from other researchers and different labs for research.

If the sequence isn't too long I PCR it from cDNA, it's often faster than the company's delivery time. Then I TOPO-clone the PCR product.

OriGene has several validated plasmid. The ones I ordered all worked very well in my hands while I had troubles with several other plasmid sources!

InVitrogen, Thermo-Scientific, Genscript

I had actually a very bad experience with Invitrogen. They send me, instead of a single plasmid, kind of a mixture of several different plasmids (when we did minipreps each miniprep had a different insert). I ask for a replacement and they sent me exactly the same staff. When I complained the answer was: "it isn't our fault; we order the clones from an external source....". I would definitively advice OriGene: they are expensive but just perfect.
