I had a kit but my PCR elisa didn't answer so I want to make the buffer myself.
Elaheh, which kit have you been using? Have you tried to contact the manufacturer? Sometimes they will tell you what's in the buffers. Probably not, but worth a try? @amanda thanks alot for your answers ,do you have aprotocol that buffer composition is with pbs not tbs?
If you prefer to use PBS then just make this up and use in place of the TBS, adding detergent where appropriate. The rest of the protocol will be as written. It shouldn't make any difference though whether you use TBS or PBS.
Add to 800ml H2O-
8 g of NaCl
0.2 g KCl
1.44 g of Na2HPO4
0.25 g of KH2PO4
Mix well then ph to 7.4 using 1M HCl.
Top up H2O to 1000ml.
Autoclave to sterilise.
@amanda ,i want to use nunc protocol here attached,but the solutions are simpler than other protocol foe example hybridization buffer is just sodium buffer phosphate ,do u thik is it good?
It's worth trying, Elaheh. It does look simpler, but I've never used such a hyb buffer before. Good luck
