But in general, you can extract total proteins from culture medium by ammonium precipitation method.

My protein is Bone Morphogenetic Protein 7. It has C-terminal His, V5 and Flag tag.

I want to do western of isolated protein e with His tag specific  antibody.

After discarding the media, first wash twice with PBS so that no media would remain in cell pallet. In case of whole cell extract, you can use RIPA, NP-40 buffer. Otherwise you can use Thermo kit for cytosolic and nuclear extract.Best of luck.
