Does anyone know why these PC12 cells of one picture look a little rough on their surfaces compared to the other one? They look slightly different from usual and I can't figure out why..Humidity issue maybe? I used the same media for both pictures.
Any ideas? Thank you!

Yes, there is an evident difference between the two. Rounding of the cells happen only when you put some cold agent like cold PBS or media which has generally not attained a temperature of 37. If moisturisation issue was there then it should be into all cells as you have put all cells in same incubator. They may achieve proper morphology after sometime or you can trypsinize them and reseed. The effect is most probably due to some cold culture agent as per my experience.

It seems that cells have undergone some kind of stress, may be related to temperature issue, as said by Ajay. Reseeding with appropriate warm media should be helpful in getting them back to usual morphology.

Definitely the cells are under some kind of stress. Generally, the pre-apoptotic cells get the round shaped morphology. Also when you treat the cells with proteolytic enzymes like trypsin the cells may undergo same kind of shape changes and finally detached from the plate. As discussed above there may be various reasons for changing this kind of morphology including changes in the pH of the culture medium, besides temperature and infections.
I have worked with these cells and noticed that using trypsin to remove them from plates for passaging will cause damage after too many uses- I ended up having cells that looked very similar to the ones in the top picture. Therefore, manual scraping for long term/ multiple passaging has worked better for me, with less cell death.
