How to calculate differentiation of a tumour cell? What are the main algorithm to analyse cancer cells? Is there any procedure to involve cancer pathways to calculate the tumor progression model?

Your question(s) is/are difficult to understand.
The differentiation state / stage of cancer cells relates to the levels of specific proteins they still express from the tissue they originate.
There are no "algorithm"(s) to analyze cancer cells per se.
In contrast, there are many algorithms to calculate cancer cell proliferation, migration, etc...
Here attached you have a movie of a human glioblastoma cell line analyzed during 72h and its 72h-visualization speeded up to a film of < 1 min. I also attached an algorith that has been developped to analyze the cancer cell migration (Debeir et al.) and an example of the biological application (Bury et al.).
