In addition to using developmental myosin, you could a BrdU assay. (e.g. http://gradworks.umi.com/14/64/1464065.html)

you can combine with myosin a fluorescent method for DNA and RNA : acridine orange.

DNA is green fluorochrome and  RNA is red fluorochrome.

Have you looked at some of the papers from the EU project RISE? There was a lot of protein work there, from Ugo Carraro, Reginald Bittner and others. The project dealt with denervated muscle, and how it regenerates under electrical stimulation. Project coordinator was Winfried Mayr.

How long after degeneration are you willing to look at these muscles? 2 days or 2 weeks or 2 months makes a huge difference!

The easiest way, it to stain nuclei with DAPI or Hoechst and look for centrally nucleated fibers.

As already suggested, you can use an anti embryonic myosin antibody, such as BF-G6 (Schiaffino et al., Exp Cell Res 163 (1): 211-220 (1986), PubMed ID 3002828) or BF-45 (Schiaffino et al., Dev Biol. 127 (1): 1-11 (1988), PubMed ID 3282936). These antibodies can be purchased from DSHB, I can send you the protocol, if you wish.

The number of centrally nucleated muscle fibers in cross sections can also be used to quantify myogenic regeneration. The number stays elevated for several weeks (some say longer) after degeneration and regeneration has taken place.

I asked that question just to better understand what Hong Huang was looking for... 2 months after it's nearly impossible to recognize which area of the injured underwent regeneration. However, 2 weeks after degeneration, it's very easy to discriminate between regenerating and non regenerating areas. First of all, centrally nucleated fibers are still visible, as Joseph said. Also the embryonic myosin is still detectable, even if it is produced just in the first few days of regeneration. Remember that myosin proteins have a long half-life (and so turnover is slow).
