I've been struggling to get transgenic tomato using two different selections: basta and hygromycin. I did test to check the minimum concentration of both and got 10 mg/l for hygromycin and 0.5 mg/l for PPT (basta). This is the minimum concentration that killed non-transformed explants on plates. However regeneration turned out to be very low and some emerging shoots are aberrant in that they are "blind shoots" that do not have apical meristem. Then I omitted selection altogether, I got PCR-positive shoots, but they are abnormal and didn't develop. I use zeatin 1 mg/l and IAA 0.1 mg/l in regeneration media.
