I'm doing a proposal for a research to identify the target and mode of action of one drug against P. falciparum using OMICS and one of the strategies is to create a resistant strain from a sensitive one against that drug and check where the mutations occurred on the genes using omics and would like some advice about recommended techniques and protocols.

If you know on the behavioral or other levels how the resistant mice differ from the sensitive mice then you can start your strains from inbreeding after which- when you have enough with families you change to outbreeding. In my papers you find several mice strains e.g. a paper on life spans of aggressive and nonaggressive strains respectively.

Do you suggest to inject the mice with Plasomodium. flaciparum (malarial parasite) and then give the mice the drug? because I want to create a resistant strain of P. falciparum from sensitive one against a chosen drug and then check the mutation responsible for the drug resistant using omics. I did not understand you correctly first, sorry. I suppose you could have 40 mice and inject them with P. Flaciparum and then give the drug. Based on the outcome start from there. My first thought was to create two strains of mice one naturally more resistent to the malarial parasite and the other not - based on the murine immune system. The rodent strains I have selected are based on genetics expressed in different behaviors but the selctions are also followed by different changes in hormon levels and the like so I would think it would be possible to start from that perspective and apply it to your needs.
