in the literature, in order to calculate this parameter , they use slide images with 20 microscopic magnification , but in our lab , we don't have this kind of microscope , can I use 10 magnification ?

Just try it with the magnification 10 and see what happens...
(Perhaps study first a (simple) system that has been described in the literature using magnification 20?)
