I am actually working on some autoimmune diseases, where I am planning to screen patients for adenosine deaminase levels. When I referred to literature, it said I need to follow Giusti Method, but I could not find the detailed protocol or any explanation of Giusti Method.
Are you interested in measuring the ADA activity or protein levels? There are commercially available ADA activity kits, such as http://www.bqkits.com/shopcart/index.php?c=0&p=48. I have used these for measuring ADA levels in synovial fluid and it has worked well in my hands. There are also ELISA's for ADA but they are pretty expensive.

I mean what is the chemical that is measured at 550nm?
It is indirectly measuring inosine by converting it to hypoxanthine and through a series of reactions converting it to a quinone dye that you read at 550 nm.
