It's fairly easy to isolate the mitochondrias (differential centrifugation on sucrose buffer or kit) from your cells and compare the cytosol vs mitochondrial localization of CytoC by western blot. Fast and easy.

I agree with Dr Khan, cytochrome c is not released out of the cell. So do some Immunofluorscence assay to detect cyto c release from mitochondria useing anti-cyto c antibody, in apoptotic conditions.
