My drug shows anti-cancer activity at as low as 5ug/ml whereas literature available shows activity around 200 to 1000ug/ml. Any possible reasons? I am doing MTT to test drug efficacy of my drug. No available literature shows activity of the same drug at such a low concentration. Should i repeat the experiments or should i change anything in my protocol? I am using 5, 10, 25, 50, 100, 200 ug/ml range of concentrations of drug for 24 hrs incubation followed by 0.5 mg/ml MTT incubation for 3 hrs.

1. Try again to check the concentration of chemotherapy drugs. Consider the volume of the culture medium.2. Check the composition of the culture medium.3. Check all components for the MTT test.
