I have rats lesioned with 6-ohda in the medial forebrain bundle, that degrades dopaminergic projection neurons to the striatum. I would like to quantify the expression on D2 receptor on the striatum and compair control and lesioned side. the problem is that on the lesioned side there are no more D2 pre synaptic receptors, and therefore I cannot quantify changes in the expression after the lesion...anybody had a similar problem and could give me a suggestion? or knows a good anti D2 antibody?

I imagine you want to quantify post-synaptic D2-family receptors on medium spiny neurons in striatum after 6ohda lesion. Immunostaining is known to be tricky/unsuccessful for dopamine receptors. One good alternative solution is to do high-resolution in situ hybiridization with either fluorescenct oligos or branched-amplification for single RNA molecule detection (both are compatible with multicolor immunolabelling). I do not recommend WB of whole tissue because pre and post-synaptic controbutions become tangled. best f
