Besides formaldehyde can anybody please suggest me which type of fumigants we can use in plant tissue culture lab? we are thinking of using gluteraldehyde based solution "Incidur" but we don't know its effectiveness in lab and in tissue cultured plants. Please share some experience or suggestions regarding the use of incidur.
Generally we used formalin for fumigation in the Tissue culture lab and the Laminar flow.Keep some formalin in a patri dish . Keep for over night. Close the lab.and in the morning remove the formalin plate covering the eyes nose with some towel.Close the door of the lab.and run fan. Enter after 1 hour.
Thank you ma'am for your kind reply. we are already using formalin in our lab. but recently there is issue of contamination so we are trying to use other fumigants. so we came across this new fumigant Incidur. we are thinking of using it once in a week. but it is gluteraldehyde based solution therefore want to know that whether this will be safe for the in vitro plants or not.
If you are trying to disinfect plant tissues, forget glutaraldehyde, it will permeate and cross-link them permanently! It can disinfect dense solid surfaces, but it will penetrate anything porous and then pose a respiratory hazard to workers (it may crosslink proteins such as cardiac muscle and trigger irreversible autoimmunity). It's not as volatile as formaldehyde, hence much more difficult to flush away.
ClO2 is used as a tissue-friendly bactericide in consumer products such as mouthwash (is your problem bacterial, fungal, or viral?) but I don't know what it does to viable plant tissues. Other chlorine-based oxidants (ClO, for example) would harm plant tissues on direct contact, as well as generate noxious chlorocarbon fumes, but they generally sterilize surfaces cheaply (the stronger iodophores are recommended for some human pathogenic viruses, however).
Thank you Riel sir for this suggestion. we just want to use that glutaraldehyde for fumigation purpose. but if it is so harmful then i think we should not use this solution in tissue culture lab.
You should note where from contamination is starting ? from media or from micro plant or explant? if you are getting infection from medium then you should increase some more pressure in autoclave and keep media at least 25 minutes inside autoclave if after sub culturing micro plant is getting contaminated then you should follow Dr. Vivek murali's advice .............
