I am working on Tagetes spp(Marigold) tissue culture. Delayed rooting observed in liquid media containing 0.5 mg/l IBA with half MS compared to solid media having same composition. Although number of roots, growth more in liquid culture rooting was delayed (almost 7 days). Can anybody have an idea/experiance behind the delayed rooting in liquid culture?
It is water stress which can stimulate the production of root rather than foliage. In liquid culture there is an absent of periodic stress which would signal the need for root allocation. In liquid culture while water would not be limited oxygenation could become limiting. The use of solid media may be able to create both more oxygenation and periodic stress which would be more optimal for rooting. Think of the growth regulation being a consequence of the plant response to the environment.
I believe that in liquid culture mediums ethylene accumulation is more than solid.
as ethylene remains dual effect, its expected than if ethylene increase more than a specific amount this cause reduction of root formation..
I believe in long-term cultures, rooting rate will diminish and will be stop at the end of culture.
subsequent subculture seems to help better root growth.
