I am having the map of the pDC515 plasmid for which I want to know the sequence. I also want to know that how the sequence of a cloned fragment in this vector can be done, if one doesn't have the required primers, as the map with me is not having any specific sequences.

I don't have the map for the requested plasmid but you can design primers about 200 bp inside the clone sequence (both sides) and get it sequenced if you don't get the map. OR contact Dr. Stephen E Moss (University College London) for map.

If you don't have the primer sequence to use in the sequencing reactions, it is best to clone your PCR product into commercially available cloning vectors like PJET cloning vector. Once you successfully clone the PCR product and confirm with restriction digestion that the insert is present in the PJET vector, you can then send the cloned plasmid for sequencing using the kit provided PJET forward and reverse sequencing primers
