Can any body tell me the best way to transfect spermatogonial stem cells of mice? I am using lipofectamine 2000, but efficiency is very low?

Transfection efficiency of lipofectamine is between 15-25% which is very low. plz suggest some more efficient method to tansfect sscs of mouse.

I think the most efficient method in this case is viral transduction, especially with lentivirus. This method was used to transduce spermatogonial cells in vitro and in vivo for transgenesis, with great sucess. Check out this review: http://www.ncbi.nlm.nih.gov/pubmed/15672832
