Dear Sir/Madam, I have started some work in wheat pollen under stress conditions. I followed a recent protocol "in vitro germination medium for recalcitrant bread wheat". But I observed coiling in pollen tubes instead of straight tubes on in vitro medium which made the measurement of tube length difficult. Straight pollen tubes provide better insertion through stigma which is naturally observed. I am using PEG-6000 in media as osmoticum and also varied its concentration. But coiling problem was not resolved.

The other problem is use of gelrite instead of agar as PEG media did not solidify with agar at pH 5.8 and Gelrite (0.2%) make the media harder.
