I have extracted a protein sequence from genbank whose starting amino acid is not methionine. Is it feasible for translation? if we need to construct the same in a vector what can we do?
You need to think about the origin of this sequence. Clearly the protein when synthesised has methionine, so you have some missing sequence. Do you have a deduced protein sequence? The question is how much is missing and what would you need to add upstream to express the authentic protein? Are there related sequences which might provide some clues?

It is common to observe an expressed protein to be without the initiating methionine.
N-terminal sequence analysis of PVDF bound expressed proteins are often found to be with and without the initiating methionine.
